I have an inexact answer for you.

In my experience, asynchronous acquisition (interrupt driven) is pretty 
performant.  You should be able to get the card's max KS/s rate.  Comedi 
is no heavier as a driver than I would imagine the NI drivers as being. 
But I have no metrics to prove this -- other than the anecdotal evidence 
that I have gotten the card's advertised max rate using comedi on several 
different E-series boards.
