You can only run one command simultaneously unless you use use the new (6 month old) feature that lets you open subdevices individually (using the /dev/comediX_subdY device files created by UDEV). Then you can run simultaneous commands on different subdevices. I don't know which drivers support this mode of operation. It's best to direct questions about that to Frank Mori Hess.
